607 Skin aging: Investigation of a synergistic effect between cigarette smoke and sun rays using tissue-engineered skin substitutes

Skin aging is influenced by two distinct processes: intrinsic and extrinsic aging. The latter regroups the external factors influencing skin aging, including cigarette smoke sun rays. While these factors, studied separately, are known to cause premature effects of their synergy have been poorly characterized. objective this project assess harmful impact on Healthy substitutes were produced according self-assembly method then exposed extract (CSE), followed irradiations at different doses UVA (5, 10 20 kJ/m2). CSE was obtained capturing using an AGI-30 impinger. analyzed histology, immunofluorescence, dot blot (collagen-I, collagen-III, collagen-IV, elastin) western (p38 MAPK, ERK1/2, JNK). A decrease in living epidermis thickness observed following exposure combined with kJ/m2 UVA. significant decreased collagen-III collagen-IV expression immunostainings when irradiated, which correlates a pro-collagen synthesis increase activity MMP-1. Finally, ERK1/2 signaling pathway seems be involved synergy. This work demonstrates synergistic effect between sunlight proteins altered during it would part caused activation pathway.
